Sampling test procedure for planktonic bacteria sampler

The planktonic sampler is a highly efficient porous suction sampler based on the Anderson impact principle with an impact velocity of 10.8m/s, which is equivalent to the fifth grade of the Anderson impact scale. Preparation of a plankton sampler before sampling

1. The temperature and humidity of the operating room (clean room) to be tested must meet the specified requirements; Static pressure difference, ventilation times and air velocity must be controlled within the prescribed value.

2. The one-way flow test should be conducted in the normal operation of the purification air conditioning system for no less than 10 minutes; The non-unidirectional flow will start no less than 30 minutes later.

3. During the static test, no more than 2 people should be in the room. The tester should be on the downwind side of the sampling port during sampling.

4, the test personnel should follow the aseptic operation procedures. Take all measures to prevent contamination of sampling orifice and other man-made contamination of samples.

5. Test personnel must wear clean clothes in line with the cleanliness level.

6. Before the planktonic bacteria test, the operating room (clean room) to be tested has been disinfected.

Sampling steps of plankton sampler

1. Petri dishes (XX clean room, XX measuring point) were prepared in a clean room with a platform. Before operation, medical defatted cotton soaked in 75% ethanol was applied to wipe the platform. Testers' hands are rigorously disinfected in the same way. In the preparation process carefully check the quality of the medium plate again, there is deterioration, damage or pollution can not be used.

2. Wipe the sampling mouth of the sampler, the sampling tube, the neck cover of the sampler, the turntable and the inner and outer surfaces of the cover with medical degreased cotton soaked in 75% ethanol.

3. After the sampler is disinfected, do not put it in the petri dish, and start the instrument to run idling for no less than 5 minutes, so that the residual disinfectant in the instrument will evaporate.

4. Set the sampling amount according to the cleanliness level of the operating room. The flow rate of the sampler (L/min)X sampling time (min)= sampling amount (L)

5. Put in the petri dish, cover the lid and adjust the gap height of the sampler. During sampling operation, the petri dish should be handled with care and the outer diameter of the petri dish should be held with "five fingers" to avoid man-made contamination of the sample as far as possible.

6. Layout and number of sampling points.

What are the key points to note when using the planktonic sampler test?

1. The instrument must be calibrated regularly according to the calibration period of the test instrument. Use instruments that have been validated and are within the validity period of use.

2. If necessary, clean the surface of the test instrument before it enters the measured area, or prepare and store it in the corresponding clean room (protect the instrument with a protective cover or other appropriate cover).

3, in the 100 level clean room with paper, the above should be covered with a transparent dust-free cover, in the 100 level clean room can not use pencil and rubber.

4, the use of the test instrument should be strictly in accordance with the operation instructions.
